Applications of BBa_I737020

Beta-Agarase is useful for lithography and agar cutting by bacteria. Beta-Agarase is an extracellular enzyme that cleaves agar into its component subunits. Agar is a pretty good bio-material, it is translucent and at higher concentrations (3-4%) is quite solid. The idea is to have a very thin layer of top agar (LB+Agar) overtop a agar substrate (Agar only). An overnight of the agarase expressing bacteria is mixed into the top agar before it is poured, and the plate is incubated overnight. The next day, agarase is induced by whatever system is present and the agarase quickly digests through the top agar and then begins to digest the agar substrate. The rate of diffusion of beta-agarase in agar is fairly low at 1% (based on Pseudoalteromonas atlantica T6C results), and is likely minimal at higher concentrations. This would allow targeted vertical digestion of agar.
